Not normal for Sassea behavior.

Sassea, me and my wife live upstairs in my mother Pat's house. Pat has a big Tiger / Maine Coon named Kit Kat that is a big baby and since it was his house first we keep Sassea upstairs with us. She gets to come down in morning when I get her breakfast. She will explore and then race me back up to so she can eat. Three days ago she began to dart out through door that keeps her upstairs when wife or I would come or go. She also began yowling to be let out of our portion of the house. For almost two years Sassea has been happy with the area we live in. She used to stop if she headed through the door and we said her name or NO. Now she quietly sasses (yat yat) back at us and keeps going. She try's to wake us to let her out the door which she never did. When we don't respond to her pleading to open the door she will stand on her hind legs to reach and rattle the door handle. Last night when Sassea wanted out I decided to follow her. She went to downstairs living room and took a bite of a Spider plant. After a couple of minutes of her just sitting next to it I told her we need to go upstairs. She resisted some and when she got to the next end table and I insisted we go she hissed at me. I am trying to understand what she wants and also not get into any habit of arguing back and forth that we're not going downstairs.

Is there any reason why you have chosen not to properly introduce the cats?  It does take some time and patience, but most cats can learn to coexist with each other.  Is she confined to one small room?  I assume since she has been downstairs to eat that she has decided there is more to her world that just an upstairs space.  It is natural for cats to be curious.  At night she might feel safest and want to explore. 

I would suggest that you look into properly introducing the cats so they each can roam the house.  I can include a few articles as well as a video on cat to cat intros.  If you take the process really slow, all should work well.

Is there any chance that your cat needs a vet visit?  Has she been spayed?  Sometimes when cats change their behavior they need a vet check up to be sure all is well.

Sassea has been spayed. She visited the vet three weeks ago for a shot, this behavior did not exist then so did not have concern at that time. Pat babies Kit Kat so to avoid antagonizing him we keep Sassea upstairs with us. She never tried to wake us to open the door like now. She lets us sleep till the breakfast alarm goes off in the morning  I understand her need to explore but the persistence is something new. Since she wanted the spider plant I figure I would take her outside this weekend. She used to go out more but now she does not like the harness so the only time she has been out lately was after her vet visit.

We found that troubling behavior went away when we took the Spider Plant away that she had been sneaking off to eat.
