Just an update, Brain did show improvement over the next day and night and he's doing much better now. Looks to be back to his normal self :) I've adjusted his diet to have more wet food and we've bought a water fountain to encourage him to drink more. Thanks to those who gave advice and comfort.

No, he wasn't able to take a sample because apparently Brain's bladder had released in the carrier on the way there so it was empty. That fits with the vet's conclusion that there's no blockage but once we got back home he started having trouble again, so I don't know what to make of that.
